Before the move to pick off a base runner, the pitcher becomes set then makes his move to the base he wants to pick the runner. Does the back leg of the pitcher need to be locked, kept bent, or does it matter before the pick off occurs?

Doesn't matter, although he can't buckle the knee or drop his heel before the step & throw.

he also can pick before he comes set or for that matter anytime before he begins his delivery.
